EUROPRISE: un réseau d’excellence européen pour de nouvelles approches sur la prévention du sida
ユーロプライズ: エイズ予防の新たなアプローチのための卓越したヨーロッパ研究拠点ネットワーク
Des chercheurs de 32 institutions (université, instituts de recherche gouvernementaux, firmes pharmaceutiques…), dont l’Institut Pasteur - situées dans 10 pays viennent de se regrouper au sein du réseau EUROPRISE (European HIV Enterprise), financé à hauteur de 15,5 millions d’euros par la Commission Européenne. EUROPRISE a essentiellement pour objectif de promouvoir le développement de candidats-vaccins et celui de microbicides, à travers le lancement d’essais cliniques dans les cinq prochaines années. Ce réseau est coordonné par le St George’s University de Londres en Grande-Bretagne (voir le communiqué ci-joint).

HIV予防の研究推進に1,550万ユーロのEUROPRISE
A €15.5 million grant from the European Commission will bring together HIV/AIDS researchers at 32 institutions - including universities, governmental research institutes and pharmaceutical industries - in 10 countries to prevent the spread of AIDS. The award holds out the promise of the most important advance yet in scientific efforts at a European-wide level in HIV-1 prevention and represents another resounding endorsement of the high commitment to HIV/AIDS prevention research in Europe.
AIDSの蔓延予防のために欧州委員会から1,550万ユーロの研究費が降り、10 カ国の大学、政府研究機関、製薬企業を含む32の研究施設のHIV/AIDS研究者を結集することになる。このグラントは、ヨーロッパのHIV-1予防研究の最も重要な進展を保証し、ヨーロッパにおけるHIV/AIDSの予防研究に深く関与することを改めて示すものである。
The European HIV Enterprise (EUROPRISE) consortium will promote an integrated program of research, coordinating a wide portfolio of activities encompassing the whole pipeline of vaccine and microbicide development from discovery through to early clinical trials for the next five years. Together these projects represent more than 150 institutions from 22 countries including two major pharmaceutical companies (Novartis and GlaxoSmith Kline). This unique approach places the network at the international forefront of understanding the interface between these two technologies (microbicides and vaccines), pursuing a critical path to the development of effective HIV-1 prevention strategies.
ユーロプライズ(EUROPRISE: European HIV Enterprise)コンソーシアムは、総合的研究プログラムを推進するもので、今後5年間でワクチンと殺ウイルス剤の発見から初期臨床試験を目指すすべての研究活動を調整する。これらのプロジェクトは22カ国150の研究施設に及び、その中にはノバルティスとグラクソ・スミスクラインという二大製薬会社も含まれている。この素晴らしいアプローチは、殺ウイルス剤とワクチンという二つのテクノロジーの共有領域を解明する国際的な最前線にネットワークを置き、HIV-1の有効な予防戦略の開発を目指している。
The urgent need for HIV prevention
HIV予防の緊急性
The UNAIDS/WHO estimates that at the end of 2004, forty million people globally were living with HIV, of which 740,000 were in Western Europe and 25 million were in Sub-Saharan Africa. Globally, there are 14,000 new HIV infections per day, 80% of which are now heterosexual, and 95% of which are in developing countries. While public health programmes encouraging the use of condoms have achieved results in reducing HIV infection rates, it is clear that additional preventative strategies are needed. Although therapeutics for HIV/AIDS continue to improve, ultimately the development of safe and effective strategies of blocking and preventing HIV transmission will be key to combating this pandemic.

Why Microbicides and Vaccines?
なぜ殺ウイルス剤とワクチンなのか
It is currently understood that conventional vaccine approaches fail to induce sufficient mucosal response and immunological memory to provide protection against the diversity of circulating HIV strains. In contrast, while it may be technically easier to develop microbicides (vaginal formulation, gels or creams) that prevent HIV transmission, their duration of protection is likely to be short lived and their efficacy will be critically dependent upon consistent use. In the past, both fields have been slow to work together in the development of products that provide multiple levels of protection. This network is focused on the premise that microbicides and vaccines that target multiple stages of mucosal transmission will have the best chance of success. Since both target the same processes there is clear synergy between the two fields.
古典的なワクチン療法は血中を循環する多様なHIV株を防御するのに充分な粘膜反応や免疫学的記憶を誘導しない、と現在では理解されている。これに対して、HIVの伝播を予防する殺ウイルス剤(ジェルやクリームの膣用仕様)の開発は技術的にはより容易であるが、予防効果を示す期間が短く、その効果は持続的な使用にかかっている。これまでこの二つの分野が多様な予防効果を示す製品の開発に共同で当たるということは少なかった。今回のネットワークの前提となっているのは、粘膜を介する伝播過程のいくつかの段階を標的とした殺ウイルス剤とワクチンが最も効果的な結果を生み出すということである。この両者が同じ過程を標的にしているので、二つの分野間で明らかな相乗効果が生れる。
What is the timescale and who is involved
今後の予定と参加者
Robin Shattock and St George’s, University of London will coordinate the consortium, together with the scientific coordination of Hans Wigzell from the Karolinska Institute (Stockholm) and Rino Rappuoli, the coordinator for HIV vaccines, from Novartis (Siena). In addition to UK, Italy andSweden, the other countries involved are Austria, Belgium, France, Germany, Netherlands, Russia and Spain.
This is a five-year project that will start on January 1st 2007 and is designed to take this approach into early human trials.
ロビン・シャトックとロンドン大学聖ジョージ医学校が、カロリンスカ研究所(ストックホルム)のハンス・ウィグツェルとノバルティス(シエナ)のHIVワクチン調整担当のリノ・ラップオリの科学的協力を得てコンソーシアムを調整する。英国、イタリア、スウェーデンに加え、オーストリア、ベルギー、フランス、ドイツ、オランダ、ロシア、スペインが参加する。
5年計画となる本研究は2007年1月に開始され、初期の臨床試験を予定している。
